With the development of next generation sequencing (NGS) technology, the cost of sequencing continues to decrease. 2. Probes & Streptavidin Magnetic Beads
Compared with solid-phase hybridization, liquid-phase hybridization has higher efficiency, simpler operation, low requirements for equipment and easy automation. The target sequence enrichment and library construction can be completed within a fer hours. Liquid-phase hybridization is considered an ideal choice for clinical testing.
Using biotin-labeled oligos as probes to hybridize with the DNA library, the target gene fragments are hybridized to the probes; and then the probes are adsorbed to the magnetic beads through the combination of biotin and streptavidin. The DNA fragments in the non-target area are washed away to obtain the target gene, and then the target area is sequenced to obtain the corresponding biological information.
